Monosodium glutamate (MSG, INS 621 / E621) supplies glutamate and normally forms the main umami foundation in a savory formulation. Disodium 5′-ribonucleotides (I+G, INS 635 / E635) is a nucleotide flavor-enhancer system commonly based on disodium inosinate (IMP) and disodium guanylate (GMP). The two materials do not perform the same job and should not be compared kilogram for kilogram.

IMP and GMP can intensify the sensory response to glutamate. This is why a small addition of a nucleotide blend may change perceived savory impact more than an equal-cost increase in MSG alone. The response is nonlinear, however, and the economically useful point depends on the complete recipe rather than a universal MSG-to-I+G ratio.

No Universal Ratio

A formula containing meat extract, tomato, mushroom, hydrolyzed vegetable protein or yeast extract already brings naturally occurring glutamate and, in some cases, nucleotide background. Salt, sweetness, acidity, fat, aroma compounds and thermal processing also change the way umami is perceived. A ratio copied from a different product may therefore overshoot, underperform or produce no useful cost benefit.

Do not assume that every commercial I+G product has the same IMP:GMP composition. Ask the supplier to state the actual composition and total-nucleotide test method. The approved specification and batch COA—not a generic product name—must define what is being purchased.

  • Map existing sources of glutamate, IMP and GMP in the base formula.
  • Keep salt, pH, fat, aroma and cooking conditions constant during the first screen.
  • Use a blank, an MSG-only control and several stepped nucleotide additions.
  • Record both immediate impact and lingering aftertaste.

Bench-Trial Design

Begin with the finished-food target, not the additive. Define whether the problem is weak broth character, insufficient meatiness, a thin middle palate, an expensive seasoning load or inconsistent impact after processing. Each objective produces a different experimental plan.

Prepare coded samples at equal salt and equal serving temperature. Compare them blind where possible. A simple triangle test can help show whether a change is perceptible, while a scored descriptive test can separate umami intensity, saltiness, aroma balance, mouthfulness and aftertaste. Repeat the best candidates in more than one batch before drawing a commercial conclusion.

  • Control: current formula or additive-free base.
  • Reference: MSG adjustment without I+G.
  • Trial series: fixed MSG with stepped I+G levels.
  • Optimization series: reduce other costly savory components only after sensory equivalence is demonstrated.

Process Validation

Evaluate the system after the real manufacturing process. Retort, baking, frying, spray drying, fermentation, freezing and long hot holding can alter the surrounding flavor matrix even when the enhancer itself remains present. A bench sample tasted before processing is not enough evidence for a commercial decision.

Recheck the product at release and again during the intended shelf life. For dry seasonings, also assess segregation, dusting, dissolution and distribution in the finished food. For liquid systems, confirm dissolution order and whether other ingredients create local concentration or precipitation problems.

Cost-in-Use

The correct commercial comparison is cost per finished batch at equivalent sensory performance—not price per kilogram of MSG or I+G. Nucleotides are usually evaluated at much lower addition levels than MSG, so quotation price alone gives a distorted picture. At the same time, more I+G is not automatically better; sensory response can show diminishing returns.

Document the approved formula, raw-material specification, addition sequence and sensory reference. This prevents a purchasing substitution from changing IMP/GMP composition, crystal behavior or flavor impact without being noticed.

Specification Checklist

A professional I+G inquiry should request more than the name E635. It should identify the IMP and GMP composition, total nucleotide result and method, moisture or loss-on-drying basis, appearance, solubility expectation, production-origin statement, allergen declarations, packaging and storage conditions. For MSG, confirm identity, assay method, crystal size, loss on drying, packing and the applicable food-grade standard.

Request a current typical specification before approval and a batch COA for shipment. Where Halal, Kosher or food-safety certification is required, verify that the document applies to the actual product and production source being quoted. Do not treat a certificate from another product or plant as equivalent evidence.

  • Exact IMP:GMP declaration and total nucleotide method
  • MSG crystal size or mesh and dissolution requirement
  • Batch COA, TDS and SDS where applicable
  • Origin, production-method and allergen declarations
  • Inner liner, outer carton or bag, net weight and storage conditions
  • Destination-market additive and labeling requirements

Regulatory Check

INS and E numbers identify additives; they do not create one worldwide permission or use level. The GSFA lists disodium 5′-ribonucleotides as INS 635 and provides international provisions, but the importing country’s current rules, food category, labeling requirements and customer standard still control commercial use.

Before scale-up, confirm the finished-food category and all relevant local restrictions with a qualified regulatory professional. Supplier documents support the review but do not replace the food manufacturer’s responsibility for formulation and labeling compliance.
